Vanderbilt-Ingram Cancer Center researchers have identified a new population of intestinal stem cells that may hold clues to the origin of colorectal cancer.
This new stem cell population, reported March 30 in the journal Cell, appears to be relatively quiescent (inactive) ? in contrast to the recent discovery of intestinal stem cells that multiply rapidly ? and is marked by a protein, Lrig1, that may act as a "brake" on cell growth and proliferation.
The researchers have also developed a new and clinically relevant mouse model of colorectal cancer that investigators can now use to better understand where and how the disease arises, as well as for probing new therapeutic targets.
Colorectal cancer is the second leading cause of cancer deaths in the United States. These tumors are thought to arise from a series of mutations in intestinal stem cells, which are long-lived, self-renewing cells that gives rise to all cell types in the intestinal tract.
For more than 30 years, scientists believed that intestinal stem cells were primarily quiescent, proliferating only rarely in order to protect the tissue against cancer. Then, in 2007, researchers reported finding a population of intestinal stem cells (marked by the molecule Lgr5) that were highly proliferative.
Those findings "really changed the way we think about intestinal stem cells," said Robert Coffey, Jr., M.D., Ingram Professor of Cancer Research, co-chair of Vanderbilt's Epithelial Biology Center and senior author on the study.
"It came to so dominate the field that it raised the question about whether quiescent stem cells even exist?and that's where we enter into the picture."
Coffey's lab studies the epidermal growth factor (EGF) signaling pathway ? which includes a family of receptors known as ErbBs ? and its role in cancers of epithelial tissues, like the intestinal tract.
Postdoctoral fellow Anne Powell, Ph.D., led the recent experiments showing that Lrig1, a molecule that regulates ErbB activity, is present in intestinal cells that have the qualities of stem cells (self-renewal, and the ability to produce all the cells of the intestine).
"Essentially, what we show is that the Lrig1-expressing cells are stem cells and they are largely quiescent," Powell said. "We also show that they're distinct from the Lgr5-expressing stem cells that had become a sort of 'hallmark' stem cell population?with different gene expression profiles and different proliferative status."
They also showed that Lrig1 is not only a marker of intestinal stem cells, but also acts as a tumor suppressor and inhibits the growth and proliferative signals of the ErbB family ? acting as a sort of "brake" on cell proliferation that can lead to cancer.
Postdoctoral fellow Yang Wang, Ph.D., eliminated Lrig1 in mice and showed that nearly all of those mice developed intestinal tumors, providing further evidence suggesting that Lrig1 functions as a tumor suppressor.
The findings underscore the importance of ErbB signaling in the behavior of intestinal stem cells from which colorectal cancer may arise.
Most exciting, said Coffey, is that the mouse model his lab has generated as a part of these studies is one of the only mouse models to develop tumors in section of the intestines where most human tumors develop: the colon. One additional advantage of this model, in contrast to others, is that the tumors develop quickly and can be easily monitored with endoscopy, which will make it easier to assess how therapeutic interventions are working.
"We are fairly confident that this will be the 'go-to' model to study colon cancer in mice for the foreseeable future," Coffey said.

WASHINGTON (AP) ? A former MF Global executive is refusing to answer lawmakers' questions about more than $1 billion in customer money that vanished in the days before the firm collapsed, invoking her Fifth Amendment right against self-incrimination.
Edith O'Brien, a former assistant treasurer at MF Global, was subpoenaed to testify before the House Financial Services oversight subcommittee hearing about an email she sent in the firm's final days.
The email appears to contradict testimony from Jon Corzine, the firm's then-CEO. It says he ordered the transfer of $200 million from a customer account on Oct. 28 to cover an overdraft in the firm's bank account in London.
Corzine testified in December that never directed anyone to use customer funds to fix the overdraft and he wasn't told that customer money was used.

Demi Lovato fans won't have to wait too much longer to see the singer's video for her latest single, "Give Your Heart a Break."
The video for Lovato's second Unbroken single will premiere April 2 on E! News. In several teased shots from the clip, a bare-faced Demi looks out from behind a curtain and flirts with her love interest. In another shot, said shirtless love interest returns her tender glances, also from behind a curtain.
"The new music video, I'm basically trying to convince a guy that I didn't break his heart, and we get into a fight and I try to win him over again," she told MTV News. "So I do something special for him at the end of the song."
The video certainly seems like it's going to be sexy, and Lovato commented on having to get romantic with someone she barely knows on set. "It's kind of funny. It's part of the job though. You just try to get through it, but it is kind of awkward at first when you have to act like a couple with someone that you don't know," she said. "He was really sweet, really sweet and really cute."
Recently, Demi opened up a bit more about the track in a video she posted on Twitter. "19-years-old ... sometimes it feels like during that brief time that I've been alive that I've lived through more emotions than people twice my age," she said. "I think that's the reason I sing about love so often in the music I make."
She explained that despite the play on the word "heartbreak" in the title, the song is about the exact opposite. "Last year I began working on a song about a different kind of love," she said. "It's a song about showing someone you love that you're the one right in front of them. This is a song about faith."
Two years ago, I loudly supported the idea that sudden-death overtime places too much emphasis on the coin flip.

Also, longer regular-season games result in more opportunities for player injuries.

"My own family and friends believe I obscured or diverted attention from the primary fact, which is that an unarmed 17-year-old was shot dead by a man who was never seriously investigated by local police. And if that is true, I apologize."
But not, he continued, because he was incorrect.
"I remain absolutely convinced of what I said about asking for trouble. There's enough trouble for minority boys not to provoke mad responses from paranoid jerk-offs."

The team found that within a microsecond of the explosion strong electron currents and magnetic fields formed around a shock wave. Astrophysicists took these results and scaled them through 22 orders-of-magnitude to find that their measurements matched the 'magnetic seeds' predicted by theoretical studies of galaxy formation.
A report of the research is published in a recent issue of the journal Nature.
'Our experiment recreates what was happening in the early Universe and shows how galactic magnetic fields might have first appeared,' said Dr Gianluca Gregori of Oxford University's Department of Physics, who led the work at Oxford. 'It opens up the exciting prospect that we will be able to explore the physics of the cosmos, stretching back billions of years, in a laser laboratory here on Earth.'
The results closely match theories which predict that tiny magnetic fields -- 'magnetic seeds' -- precede the formation of galaxies. These fields can be amplified by turbulent motions and can strongly affect the evolution of the galactic medium from its early stages.
Dr Gregori said: 'In the future, we plan to use the largest lasers in the world, such as the National Ignition Facility at the Lawrence Livermore National Laboratory in California (USA), to study the evolution of cosmic plasma.'

Good agricultural practices -- GAPs -- are food-safety standards that reduce the risk of on-farm produce contamination, according to Daniel Tobin, a doctoral candidate in Agricultural and Extension Education and the principal investigator in the study. GAPs include recommendations for safe irrigation methods, use of raw and composted animal manure, worker health and hygiene, post-harvest handling practices and traceability procedures.
On-farm, food-safety measures are critical, Tobin noted. The U.S. Centers for Disease Control and Prevention estimates 48 million foodborne illnesses, including 3,000 deaths, occur each year in the United States. Previous research indicates that there is a rising number of reported outbreaks in fresh produce.
For Pennsylvania produce growers to maintain wholesale market opportunities, they will have to put forth substantial effort to comply with and verify their on-farm, food-safety practices, the researchers reported in recent issues of Food Control and the Journal of Extension.
"Growers, therefore, will need GAP training and educational materials," Tobin said. "General training about potential on-farm, food-safety hazards and preventative measures no longer will be adequate. To achieve maximum impact, the curriculum must focus on implementing and documenting GAPs so growers will be prepared to pass a GAP audit.
"In response, supermarkets have been implementing policies that require their fresh produce suppliers to attend training workshops on farm food safety and/or to verify compliance with food-safety standards through fee-based, third-party certification. In TPC an independent, third-party auditor conducts an on-site inspection to determine whether a supplier's practices and procedures comply with a certain set of standards, such as GAPs."
Produce growers who need to verify GAP conformity through third-party compliance can select among several audit agencies. The U.S. Department of Agriculture's Agricultural Marketing Service, in association with state departments of agriculture, offers a voluntary audit program that growers can use to verify on-farm food-safety practices. Private companies also offer audit services, but their GAP standards and documentation requirements can vary from company to company.
The Food Safety Modernization Act, signed into law in early 2011 by President Obama, mandates the Food and Drug Administration to establish mandatory minimum standards, based on known safety risks, for produce growers to implement and document.
To help develop a relevant GAP training curriculum that meets the needs of Pennsylvania growers, Penn State researchers surveyed supermarkets that operate in the state to determine their food-safety policies and practices that affect local produce growers.
Pennsylvania consumers also were surveyed because they help drive demand for produce. The researchers found that consumers are highly concerned about the safety of the produce supply.
"Assessing American consumer perceptions regarding produce safety is particularly important, because those perceptions will allow actors within the supply chain to better meet consumer demand," Tobin said. "Using data collected from Pennsylvania consumers, our study documents how consumer demographics -- and their preferences for specific attributes in fresh produce, such as local, organic and inspected for food safety -- affect their produce-safety perceptions."
Such an analysis provides important information for those seeking to implement practices that reduce the risk of foodborne contamination, Tobin explained. A clearer understanding of consumer produce-safety perceptions and preferences will allow these groups, including growers and supermarkets, to make better-informed decisions regarding their food-safety policies and practices.
